Biocompatible Nanoparticles as a Platform for Enhancing Antitumor Efficacy of Cisplatin–Tetradrine Combination

Abstract Combination therapy has been a standard strategy in the clinical tumor treatment. We have demonstrated that combination of Tetradrine (Tet) and Cisplatin (CDDP) presented marked synergistic anticancer activity, but inevitable side effects limit their therapeutic concentration. Considering different physicochemical pharmacokinetic properties two drugs, we loaded them into nanovehicle together by improved double emulsion method. The nanoparticles (NPs) were prepared from mixture poly(ethyleneglycol)–polycaprolactone (PEG–PCL) polycarprolactone (HO-PCL), so CDDP Tet can be located NPs simultaneously, resulting low interfering effect high stability. Images fluorescence microscope revealed cellular uptake both hydrophilic hydrophobic agents delivered NPs. In vitro studies on cell lines tissue increased inhibition apoptosis rates. As to vivo studies, superior antitumor efficacy reduced observed group. Furthermore, 18 FDG-PET/CT imaging metabolic activities tumors more prominently. Our results suggest PEG–PCL block copolymeric could promising carrier for combined chemotherapy with solid minor effects.
